Description / Use

Irvine Scientific’s Vit Kit® – Freeze and Vit Kit® – Thaw consist of solutions optimized for vitrification and recovery of oocytes (MII stage)
and embryos (zygote to blastocyst stage).

Media Composition

The Composition of these solutions is based upon publish methods utilizing a combination of ethylene glycol (EG) and dimethylsulfoxide (DMSO) as the permeating cryoprotectants. The basal medium for these solutions consists of Modified M199 (containing 21mM HEPES buffer) supplemented with 20% DSS(HSA and Dextran, containing 10 mg/mL protein) and 35ug/mL gentamicin as the antibiotic.

Polyvinylpyrrolidone (PVP) With HSA

Description / Use

Ployvinylpyrrolidone (PVP-K90) is a synthetic copolymer with a molecular weight of 360,000 Daltons. PVP is intended for use in ICSI procedures involving immobilization of sperm.

PVP with HSA solution is a ready-to-use PVP with HSA (5mg/mL), reconstituted in an isotonic HEPES buffered HTF medium.

7% PVP With HSA Solution

Recommended for use with low quality or low motility sperm specimens.
Available in kits of 5×0.5mL.
Has a shelf-life of 6 months from the date of manufacture when stored at the recommended temperature of 2℃ to 8℃

10% PVP With HSA Solution

Recommended for use with highly motile sperm specimens.
Available in kits of 5×0.5mL.

Lyophilized PVP* is composed of USP grade PVP which is dissolved in Water for Injection (WFI) grade water and then membrane (SAL10) filtered and lyophilized.

10% Lyophilized PVP

Available in kits of 10x1mL.
Requires aseptic reconstitution with an appropriate medium (such as sperm washing medium) prior to use.
Has a shelf life of 2 years from the date of manufacture, once reconstitute lyophilized PVP mau be stored for up to 3 weeks when stored at the recommenced temperature of 2℃p to 8℃.

Isolate® Sperm Separation

Description / Use

Isolate® is a superior medium for separation of the motile fraction of sperm from its surrounding seminal plasma by density centrifugation.

As a two-layer gradient system, ISolate effectively removes cellular contaminants-dead sperm, white cells, and miscellaneous debris-yielding a sample that contains predominately motile sperm.

ISolate is a sterile, colloidal suspension of silica particles stabilized with covalently bound hydrophilic silane is a HEPES-buffered HTF. This buffered system maintains a physiological pH at ambient conditions (do not use this product in a CO₂ incubator unless tightly capped).

a fresh or cryopreserved semen sample is overlaid on 1.5 to 2.0mL aliquots of the two-layer gradient system in a conical centrifuge tube. The prep tube is then centrifuged at a low speed, ~300xg for 20 minutes. This action causes seperation of the seminal components. Dead sperm and debris are found in the upper portion of the tube while a fraction of highly motile, morphologically normal cells will form a pellet in the bottom.

Irvine Scientific has conducted extensive performance and safety testing on ISolate. These studies indicate that it is comparable or superior to the standard separation techniques utilizing Percoll® and superior in performance to standard swing-up procedures.
